Een partner gaat uiteraard geen miljoenen in TiGenix pompen zolang het EMA geen vergunning heeft verstrekt voor het in de handel brengen van Cx601. Die vergunning is inmiddels aangevraagd. Zodra die vergunning is verleend staan alle seinen op groen. Tot die tijd uiteraard geen IPO. Als het allemaal loopt zoals ik denk dat het zal lopen zitten er tientallen procenten koerswinst aan te komen.

Dan zeg je dus eigenlijk dat de IPO nog zo'n 300 dagen op zich laat wachten.

Tigenix gaat binnen 1 maand antwoord krijgen of hun dossier aanvaard is. Vanaf dan loopt dag 1. Na 120 dagen krijgt tigenix een loq (list of questions) waarvoor ze 60 dagen hebben om te antwoorden. Tijdens die periode list de klok stil. Nadat tigenix op loq heeft geantwoord loopt de klok opnieuw tot dag 180 waarop tigenix een LoOI: (List of Outstanding Issues) krijgt. Ze hebben hier 1 maand de tijd om te antwoorden. Ook dan ligt de klok stil. Na antwoord hierop loopt de klok tot dag 210 waarop de CHMP zijn advies geeft aan de EMA. De EMA beslist dan tegen dag 277 of ze definitief goedkeuring geven. Meestal volgt de EMA het advies van de CHMP.

Waarom zou tigenix geen partner kunnen vinden zolang de goedkeuring er niet is? Dit gebeurt regelmatig op die manier. De kans op goedkeuring ligt volgens mij rond de 90%. Meestal wordt er een deel betaald voor de deal en volgen er nog betalingen bij de goedkeuring en als de eerste patiënt gedoseerd is.

De vraag is wil tigenix een deal in de EU? Als je leest op hun company presentatie spreken ze van bepaalde landen. De zogenaamde vetpotlanden (Duitsland, GB, Frankrijk, Italië, Spanje) en nog enkele andere landen willen ze zelf bedienen. De vraag is dan nog wie een deal wil voor de restjes in de EU? Dit is nooit zonder risico, nieuw product,nieuwe techniek, soms jaren wachten op goedkeuring autoriteiten voor de terugbetaling is geregeld, de lucratieve bezigheid van de chirurgen (operatie)

The filing of the dossier to gain European approval comes in-line with
previous guidelines. Tigenix reconfirmed its ambition to gain approval in 2017
allowing to bring the product to the market in 2H17.

Detailed results of the phase 3 study on which the MAA is built will be
presented within 2 weeks during the Congress of European Crohn's and
Colitis Organisation (ECCO) in Amsterdam on 17th and 18th March 2016

Tigenix is heel goed bezig. Ik houd mijn stukken stevig vast. De koers blijft redelijk stabiel staan maar dit is de stilte voor de storm. Dit aandeel kan de komende tijd nog wel een paar keer over de kop.

EMA en IPO staan los van elkaar.

Pinkie zet je roze bril op, komt allemaal goed!
Wachten is op the NASDAQ IPO. Alles is in kannen en kruiken behalve het markt omveld. Sentiment voor BioTech is nog steeds niet positief. IBB heeft ongeveer 25% verloren sinds 1.1.2016 en heeft de stijgende trend nog niet te pakken. In dat omveld een IPO te doen is niet verstandig. Ben er zeker van dat er nu gesprekken gaande zijn over de IPO prijs. En zolang die niet zijn wat men wil hebben (danwel denkt waard te zijn) zal de IPO niet van start gaan. Maar het sentiment op de beurs word langzaam beter dus het kan elk moment komen. En ik verwacht dan een behoorlijke koers sprong. De Tigenix boog is gespannen en wacht op het loslaten.......

Meer dan 1 op 2 patiënten is gebaat met de celtherapiebehandeling van het Leuvense biotechbedrijf. De beursgang op Nasdaq kan nu in een stroomversnelling komen.

Dat blijkt uit nieuwe resultaten van een grootschalige studie op patiënten die niet reageerden op andere behandelingen.

In augustus vorig jaar bleek al dat een behandeling met Cx601 zeer nuttig is. Dat is een celtherapiebehandeling van Tigenix om 'complexe perianale fistels bij patiënten met de ziekte van Crohn' te genezen. Na 24 weken was de helft van de patiënten genezen.

Nu blijkt dat de eenmalige injectie na één jaar nog altijd een positief effect heeft. Dat is niet onbelangrijk. Een dure behandeling die je om de zoveel tijd moet herhalen, heeft namelijk minder kans op terugbetaling door de ziekteverzekering. Eén jaar na de injectie is 54,2 procent van de patiënten vrij van kwaaltjes. Het verschil met de groep die een placebo toegediend kreeg is statistisch significant, ook al toonde 37,1 procent van die placebogroep een verbetering.

Tigenix spreekt in een persbericht over een 'grote doorbraak' en 'een nieuw paradigma voor de behandeling van deze chronische ziekte'.

Cx601 is het uithangbord van Tigenix. En met deze resultaten op zak, kunnen de beursgang op Nasdaq en de onderhandelingen met een farmapartner in een stroomversnelling komen. Tigenix zoekt die partner om een laatste studiefase in de VS te financieren.

-A single injection of Cx601 was statistically superior to placebo in achieving combined remission at week 52 in the treatment of complex perianal fistulas in Crohn's disease patients with inadequate response to previous therapies, including anti-TNFs
-54.2% of patients treated with Cx601 achieved combined remission at week 52 compared to 37.1% in the placebo arm
-75.0% of Cx601 treated patients who achieved combined remission at week 24 remained in combined remission at week 52 compared to only 55.9% in the placebo arm
-The results confirm the favorable safety and tolerability profile of Cx601 already
reported at week 24

Leuven (BELGIUM) – March 7, 2016, 19:00h CET – TiGenix NV (Euronext Brussels: TIG), an advanced biopharmaceutical company focused on developing and commercialising novel therapeutics from its proprietary platforms of allogeneic expanded stem cells, announced today that a single injection of Cx601 was statistically superior to placebo in achieving combined remission at week 52 in the Phase III ADMIRE-CD trial in Crohn's disease patients with complex perianal fistulas with inadequate response to previous therapies, including anti- TNFs. The one-year data also confirm the favorable safety and tolerability profile of Cx601 already reported at week 24.

ADMIRE-CD is a randomized, double-blind, placebo-controlled Phase III study designed to confirm the efficacy and safety of a single injection of Cx601 in the treatment of complex perianal fistulas in Crohn's disease patients. In total, 212 patients were randomized in 7 European countries and Israel. Patients included in this study had an inadequate response to at least one previous therapy, including anti-TNFs. Continuation of medical standard of care was allowed during the duration of the trial in both groups. The study primary endpoint was combined remission at week 24, defined as closure of all treated external openings draining at baseline despite gentle finger compression, and absence of collections >2cm confirmed by MRI1. This same endpoint of combined remission has been analyzed after 52 weeks as a secondary variable.

In the ITT2 population (n=212), Cx601 achieved statistical superiority (p=0.012) with 54.2% combined remission at week 52 compared to 37.1% in the placebo arm. In the mITT3 population (n=204), the combined remission at week 52 was 56.3% and 38.6% for Cx601 and placebo respectively (p=0.010). Efficacy results were robust and consistent across all statistical analyses.
The week 52 data also shows a higher rate of sustained closure in those patients treated with Cx601 and in combined remission at week 24 (75.0%) compared to patients in the placebo group (55.9%).

1 MRI: Magnetic Resonance Imaging
2 ITT: Intention to treat i.e. all patients randomised in the trial.
3 mITT: modified Intention to Treat i.e. all patients randomized and treated, and had at least one post-baseline efficacy evaluation.

Treatment-emergent adverse events (non-serious and serious) and discontinuations due to adverse events were comparable between Cx601 and placebo groups.
"We are delighted with the remarkable consistency of these results with respect to those reported at 24 weeks. In particular, the 75% persistence in the combined remission from week 24 to week 52 confirms Cx601 as a promising therapeutic solution for a hard to treat population that is refractory to existing treatments including anti-TNFs,” said Dr Marie Paule Richard, Chief Medical Officer of TiGenix. “The improvement brought by Cx601 compared to the best standard of care available could make a real difference to the life of patients suffering from this chronic disease”.

"The one year results of this large robust controlled study open up a new paradigm for the treatment of perianal fistulising Crohn's disease, one of the most severe manifestations of this process. With a single injection of Cx601 more than 50% of patients are in remission at one year with a favourable safety profile. Remarkably, most patients that were already in remission at six months continue to benefit from the treatment six months later, a major breakthrough in this field”, said Dr Julián Panés, Head of the Gastroenterology Department, Head of the Inflammatory Bowel Diseases Unit, and Associate Professor of Medicine at the Hospital Clínic of Barcelona, President-Elect of ECCO, and Chairman of TiGenix ADMIRE-CD Scientific Advisory Board.
