Ik las in het lokale blaadje over Elin van 9 jaar. Zij is ook het meisje uit het fimplje. Naast mijn eigen bijdrage, via de 2e link, hoop ik ook een enkeling van jullie te bereiken. Een CF medicijn van Galapagos betekent waarschijnlijk veel voor jullie pensioen, heel crue gezegd ook voor het hare.

Een van de belangrijkste redenen dat ik jaren gelden in Galapagos ben gestapt is dat ik dan met mijn bescheiden bijdrage Galapagos heb gesteund in het ontwikkelingen van medicijnen.
Uiteraard hoopte ik dat dat ze ook zou gaan lukken, het lijkt er nu sterk op dat dat gaat gebeuren.
Dat ik daar een dik rendement mee ga halen is natuurlijk wel een fijne bonus, dat was ook een reden om in te stappen.

De Universiteit van Alabama, Galapagos en AbbVie rapporteren de evaluatie van combinatie(s) van nieuwe correctoren, een potentiator en read through agents (stoffen die cellen helpen om genetische informatie met onderbrekingen toch te kunnen decoderen) om de werkzaamheid in CFTR cellen met verschillende nonsens mutaties te verhogen naar niveaus die waarschijnlijk therapeutisch effectief zijn in CF.

Poster 192 - Discovery and characterization of ABBV/GLPG2222, a novel first generation CFTR corrector
Galapagos en AbbVie rapporteren de identificatie en in vitro kenmerken van ABBV/GLPG2222, een nieuwe, krachtige en oraal biobeschikbare corrector die momenteel wordt getest in een klinische studie, en die in vitro betere effecten heeft laten zien dan bestaande correctoren in klinische studies.

Poster 252 - Safety, tolerability and pharmacokinetics of a novel CFTR corrector molecule GLPG2222 in healthy volunteers
Galapagos en AbbVie rapporteren de resultaten van de First-in-Human studie met GLPG2222. Veiligheid en verdraagbaarheid werden onderzocht en orale dosering van enkelvoudige (tot 800 mg) en meervoudige oplopende doseringen (tot eenmaal daags 600 mg gedurende 14 dagen) bij gezonde vrijwilligers. Het farmacokinetische profiel toont aan dat GLPG2222 snel werd opgenomen met een eliminatie halfwaardetijd van 12 uur; steady-state (stabiele plasma spiegels) werd bereikt na twee dagen, met een beperkte stapeling. GLPG2222 werd over het algemeen goed verdraagbaar bevonden.

Poster 253 - GLPG1837 in subjects with cystic fibrosis and the S1251N mutation: results from a phase2a study (SAPHIRA 2)
Nieuwe potentiator GLPG1837 werd toegediend in twee doseringen gedurende twee weken aan zeven patienten met de S1251N mutatie in een kleine, verkennende, open-label studie in meerdere ziekenhuizen. Het doel van de studie was om de in vitro observaties te bevestigen door klinische resultaten. De gekozen doseringen in SAPHIRA 2 waren aan de ondergens van het therapeutische spectrum, met plasmaconcentraties van GLPG1837 rond de minimale target concentratie voor werkzaamheid. Zelfs bij deze lage doseringen bleek in Kaydeco-naïeve patiënten dat de waargenomen CFTR activiteit, gemeten als de afname van chloride in transpiratievocht en als toename in FEV1 (veranderingen in absolute percentage voorspelde FEV1) ten opzichte van begincondities, in lijn was met wat is gepubliceerd over Kalydeco in S1251N patiënten na een behandeling van twee weken. Het stopzetten van een behandeling met Kalydeco had na 7 dagen een gering effect op de longfunctie (-3%), terwijl gedurende de behandeling met GLGP1837 geen verdere achteruitgang werd waargenomen bij patiënten die eerder met Kalydeco werden behandeld. GLPG1837 is over het algemeen goed verdraagbaar bevonden bij CF patiënten na een behandeling van in totaal 4 weken.

Poster 252 - Safety, tolerability and pharmacokinetics of a novel CFTR corrector molecule GLPG2222 in healthy volunteers
Galapagos and AbbVie report the results for the First-in-Human study with GLPG2222. Safety and tolerability were evaluated in oral single (up to 800 mg) and multiple ascending doses (up to 600 mg q.d. for 14 days) in healthy subjects. The pharmacokinetic profile showed GLPG2222 to be rapidly absorbed with an elimination half-life of 12 hours, steady-state after 2 days, and minimal accumulation. GLPG2222 was found to be generally well tolerated

Poster 253 - GLPG1837 in subjects with cystic fibrosis and the S1251N mutation: results from a phase2a study (SAPHIRA 2)
Novel potentiator GLPG1837 was administered in two doses, each for two weeks, to 7 patients with the S1251N mutation in a small, exploratory, open-label, multi-center study. Purpose of the study was to confirm in vitro observations with clinical responses. The doses selected for SAPHIRA 2 were at the low end of the efficacious dose range, whereby the exposures of GLPG1837 in plasma ranged around the lower target concentration for efficacy. Even at these low doses, CFTR activity was observed through decreases in sweat chloride and increases in FEV1, with changes in absolute percent predicted FEV1 from baseline in Kalydeco naïve patients being in line with those published for Kalydeco in S1251N subjects after two weeks treatment. A 7-day pre-treatment washout of Kalydeco impacted lung function slightly (-3%), and during treatment with GLPG1837, no further decline was observed in Kalydeco experienced patients. GLPG1837 was generally well tolerated in CF patients when dosed up to 4 weeks.

Heel benieuwd naar de resultaten van Safira 1. Uit het preklinisch onderzoek blijkt namelijk dat GLPG1837 vooral op mutatie G551D een (veel) betere werking laat zien dan Kalydeco.

1 op de ca. 25 Nederlanders is drager, zijn beide ouders drager dan heeft elk kind 25% kans op de ziekte.

Korte termijn koers reageert harder op wat met commerciële doeleinden door buitenstaanders wordt geroepen (analisten) dan wat Gala zelf rapporteert. Maar bevestigend nieuws van Gala is, ook als dat niets doet met de dagkoers, mij veel meer waard.

NielsjeB, ik ben het hier wel en niet met je eens.

Wel als in dat er inderdaad niet altijd op goed nieuws wordt gereageerd zoals wij hopen en vinden dat zou moeten (op de korte termijn).

En niet omdat ik denk dat goed nieuws eerst op de juiste plekken terecht moet komen, dan geïnterpreteerd moet worden en voordat men dan eindelijk op de "Koop bestens" knop gedrukt heeft kan vaak wel wat tijd tussen zitten (tijd tussen uitkomen nieuws en stijging koers / koop).
