Daggrafiek: Deze week begon met een lange positieve candle, die vanaf het 50% niveau het 76,4% bereikte, en tevens een overbought situatie bewerkstelligde. De week eindigde met een positieve candle die geheel boven het 61,8% niveau bleef. De volgende horde naar boven is nu het 76,4% niveau op 32,58. Steun mogen we verwachten van het nu bereikte 61,8% niveau.
Weekgrafiek: De candle 29,28-31,40 bevindt zich in zijn geheel boven het 76,4% niveau van de totale stijging 8,43-35,50. We mogen dus een stijging verwachten naar het 100% niveau 35,50. De snelheid van deze stijging zal bepaald worden door de stijgende maar betrekkelijk dichtbij zijnde bovenste bandbreedte rond het gemiddelde, die al snel weer een ob situatie kan veroorzaken, zoals dat ook deze week het geval was. De indicator staat positief.

Een sessie waarbij met een meer dan gemiddeld volume eerst het 76,4% niveau geslecht werd, daarna de OB weerstand op 33,26, daarna ook nog de bovenkant van het trendkanaal...33,48.
Dit was wat te veel van het goede, en daarna keerde het aandeel terug naar de ob waarde op 33,26.

Beste mensen, heb mijn geuzennaam Gaucho (werd ooit in Buenos Aires geboren) veranderd in de naam waar ik mee Twitter: DrMedicalValue.

Het is helemaal gewelidg wat Flosz en de Prof via Twitter doen en dat gaat niet ongemerkt voorbij. Er is echt een soort positieve dynamiek op Twitter aan het ontstaan rondom $QURE. De mensen daar zien nu ook wel in hoe sterk QURE ondergewaardeerd is. Chapeau voor beide!

Een marketcap van een miljard eind 2015 betekent een koers van USD 46,70. Niet ondenkbaar. De afgelopen 6-7 maanden brachten een koerstoename van ongeveer 20 USD, dus dat zou 53 USD zijn tegen einde jaar....

Wat een topdag. Nu de instapkoers van BristolMyers weer is aangetikt heb ik het gevoel dat er veel minder ankerpunten meer zijn voor de koers. Daardoor kunnen er wellicht mooie dingen gebeuren de komende tijd.

De laatste tijd kwam er na zo'n dag een suk slecht nieuws wat de koers weer terguzette, maar daar heb ik nu nog geen aanwijzingen voor. Misschien gaat het feestje nog even door. Zie de Prof zijn eventkalender.

In connection with a Collaboration and License Agreement (the “Collaboration and License Agreement”), dated April 6, 2015, that provides Bristol-Myers Squibb Company (“BMS”) with exclusive access to uniQure N.V.’s (“uniQure”) gene therapy technology platform for multiple targets for cardiovascular and potentially other diseases, BMS acquired from uniQure on June 12, 2015 1,112,319 ordinary shares, or 4.9% of uniQure’s outstanding ordinary shares immediately following such issuance (the “Initial Closing”). These shares were acquired pursuant to a Share Subscription Agreement between BMS and uniQure, dated April 6, 2015. The percentage of ordinary shares reported owned by BMS was calculated using as the denominator 22,700,390 ordinary shares outstanding as of June 12, 2015, which is comprised of 21,588,071 shares that uniQure represented to BMS was outstanding immediately prior to uniQure’s issuance of shares to BMS at the Initial Closing, plus the 1,112,319 shares issued to BMS at the Initial Closing.

After July 12, 2015, but prior to December 31, 2015, BMS is obligated to acquire a number of additional shares that, together with the shares acquired at the Initial Closing, would equal 9.9% of the outstanding shares immediately following such acquisition (the “Second Closing”). uniQure also has granted BMS two warrants (“Warrant 1” and “Warrant 2”). Pursuant to Warrant 1, BMS may acquire, in its sole discretion, an additional number of shares that, together with the shares acquired at the Initial Closing and Second Closing, would equal 14.9% of uniQure’s outstanding shares immediately after such issuance. Pursuant to Warrant 2, BMS may acquire, in its sole discretion, an additional number of shares that, together with the shares then owned by BMS, would equal 19.9% of uniQure’s outstanding shares immediately after such issuance. The exercise of each warrant is conditioned upon the designation by BMS of a certain number of additional collaboration targets and the payment by BMS to uniQure of related fees under the Collaboration and License Agreement.

uniQure and BMS also entered into an investor agreement regarding certain rights and restrictions relating to uniQure’s ordinary shares acquired or to be acquired by BMS, including (i) certain ‘‘standstill’’ restrictions that, subject to certain exceptions, restrict BMS from acquiring shares of uniQure in addition to any shares acquired in the manner described above and from taking certain other actions with respect to uniQure or its shares, its assets or its board of directors; (ii) lock-up restrictions that restrict BMS from selling or disposing of the shares acquired or to be acquired for a certain period without uniQure’s prior consent; and (iii) the requirement that BMS vote all of its shares in favor of all items on the agenda for the relevant general meeting of shareholders of uniQure as proposed on behalf of uniQure, subject to certain exceptions, including BMS’ voting rights relative to certain change of control transactions. The voting provisions will terminate upon the earlier to occur of (1) the date on which BMS no longer beneficially owns at least 4.9% of the outstanding shares of uniQure, (2) the consummation of a change of control transaction relative to uniQure and (3) the termination by BMS of the Collaboration and License Agreement for breach by uniQure.

Regelmatig omlaag op kleine omzetjes en omhoog op grotere omzetten...dat past in mijn visie op dat er wat speelt met QURE...geen gewone handel!

AMT-021 for Acute Intermittent Porphyria

We are developing AMT-021 as a gene therapy for acute intermittent porphyria, or AIP, a severe liver disorder. AMT-021 consists of an AAV5 vector carrying a therapeutic porphobilinogen deaminase, or PBGD, gene that we exclusively license from Applied Medical Research Center of the University of Navarra in Spain. Our former collaborator Digna Biotech has completed a Phase I

clinical trial of AMT-021 in eight patients in Spain. The collaboration achieved its primary goal in completing a successful Phase I study, thereby establishing the preliminary safety profile of liver-directed gene therapy with AAV5 that we expect will support future clinical studies.

Phase I Clinical Trial Sponsored by Digna Biotech

Digna Biotech conducted a multicenter, open label, prospective, interventional, single dose, dose-escalation Phase I clinical trial to investigate the safety and tolerability of AMT-021 in eight patients with severe AIP. Digna Biotech conducted this clinical trial at two sites in Spain. There were four dosing cohorts in the trial, with two patients per cohort. Digna Biotech monitored all patients for one year following treatment. The primary objective of this Phase I clinical trial was to assess the safety of systemic administration and to determine the maximum tolerated dose of AMT-021. Secondary objectives included measuring urinary levels of toxic metabolites to determine whether these metabolites can be used as a biomarker of potential treatment effect. Preliminary analysis of the one-year follow-up data indicates the safety and successful transduction of liver cells with the PBGD gene. Key findings were as follows:

· There were no liver enzyme perturbations.

· There was no evidence of cellular immune response against the AAV vector or the PBGD gene.

· Vector genomes were detected in patients’ liver biopsies obtained under one year after vector injection, indicating that AMT-021 sustainably transduced human liver cells.

Digna has advised us that there were four serious adverse events in this trial; however the events were determined by the investigator not to be treatment related. Digna further reported that there were no treatment-related adverse events or liver events related to AMT-021. Digna did not observe a reduction in the urinary levels of toxic metabolites in trial participants that might have served as a surrogate marker for efficacy. We believe that this result may suggest that a relatively high level of transgene expression will be required for a gene therapy to provide a clinical benefit in this indication. This contrasts with an indication such as hemophilia, in which the near or total absence of a protein in the patient means that a relatively low level of gene expression may result in a clinical benefit. In light of the absence of dose-limiting toxicities in the Phase I clinical trial, we are currently assessing whether higher dose levels or a more potent vector may be the relevant next step in the project. Under our consortium agreement with Digna Biotech and the other consortium members, we have an exclusive right to use all data related to the program.

Gisteren eindigde het aandeel met meer dan gemiddeld volume op de rand van OB gebied, na eerst door de 76,4% weerstand te zijn gegaan en daarna, ob zijnde toch nog de bovenkant van het trendkanaal te hebben gepasseerd. Vandaag volgde daarop als reactie, met een lager dan gemiddeld volume, een rode candle, waarbij het 76,4% niveau moest worden prijs gegeven. Bedenk wel dat het 76,4% niveau nog niet was terug getest. Het 76,4% niveau is nu opnieuw de weerstand. Als steun zie ik de onderkant van het stijgende trendkanaal.
